Use of phage display to isolate specific human monoclonal antibody fragments against a potential target for multiple myeloma.

Ann Acad Med Singapore 38(7):621-9 (2009) PMID 19652854

Multiple myeloma (MM), a malignancy of plasma cells, accounts for 10% of all haematological malignancies and is currently incurable. Although it can be treated, the disease tends to relapse after several years and becomes increasingly resistant to conventional therapy. Investigations into using humoral therapy for MM are now underway with a view that novel therapeutic agents may provide a more targeted therapy for MM.

We observed that DHA-treated trophozoites were detached from the wall of the culture tube and shown bradypraxia and bubbles in the dorsal and ventral surfaces. Ultrastructural observations by TEM revealed that DHA promoted modifications of the cell shape, pronounced dorsal vesiculation, plasma membr...

Major difficulties in phosphoprotein analysis relate to the presence of a huge number of nonphosphorylated proteins and to the wide concentration dynamic range among them. In order to overcome the analysis complexity, specific clean-up and highly efficient enrichment procedures are mandatory prior t...

We were able to identify DiART-labeled peptides with high confidence and quantify them with high accuracy. Furthermore, we adopted a hybrid electron-transfer dissociation (ETD)-HCD acquisition protocol and developed a novel data analysis approach to measure phosphorylation of peptides. Our results s...
