Aromatase inhibitors: prediction of response and nature of resistance.

Expert Opin Pharmacother 11(11):1873-87 (2010) PMID 20497094

Aromatase inhibitors (AIs) are recommended for and central to endocrine management of breast cancer patients. Response rates can be high, but resistance is a major obstacle. Optimal management therefore requires accurate prediction of response and an understanding of the nature by which resistance occurs. These are the subjects of this review.

Different dynamic patterns of expression of oestrogen-regulated and proliferation genes were observed in tumours clinically resistant to neoadjuvant letrozole, thus illustrating heterogeneity of resistance and discriminating molecular sub-classes of resistant tumours. Molecular phenotyping might hel...

Acquired resistance to either tamoxifen or aromatase inhibitors (AI) develops after prolonged treatment in a majority of hormone-responsive breast cancers. In an attempt to further elucidate mechanisms of acquired resistance to AIs, MCF-7aro cells resistant to letrozole (T+LET R), anastrozole (T+ANA...
